SSP Kits (Gel Electrophoresis)
Principle
With the use of Polymerase Chain Reaction (PCR) in HLA-typing much progress has been made recently. Sequencing of numerous alleles allows specific and clear typing on DNA level with high resolution and other decisive advantages over the traditionally used serologic methods.
The test procedure is done by using Sequence Specific Primer-(SSP)-PCR. This method is based on the fact that for a successful reaction both primers, especially at the 3' end, must not show any mismatch. Therefore only if the primers entirely correspond with the target sequence an amplification is obtained which is visualized by subsequent gel-electrophoresis. The reaction mixtures are pre-aliquoted, ready to use and thus also help to reduce handling errors.
